Upadacitinib 45mg ER Tablets

Upadacitinib 45mg ER tablet is an extended-release JAK inhibitor used for induction treatment of active ulcerative colitis or Crohn disease in eligible adults. After induction, treatment changes to a lower maintenance strength.
Swallow the extended-release tablet whole with water at a consistent time, with or without food. Do not crush, split or chew it. Avoid grapefruit and grapefruit juice, which can increase upadacitinib exposure.
Upadacitinib inhibits JAK-mediated cytokine signaling, reducing immune-driven inflammation in selected inflammatory diseases.
Possible effects include upper respiratory infection, nausea, acne, headache, fever, cough, herpes zoster, raised cholesterol, and lab changes. Serious concerns include severe infection, TB, malignancy, MACE, thrombosis, GI perforation, liver injury, low blood counts, and allergic reaction.
Tell your doctor about infection symptoms, tuberculosis or hepatitis, heart or blood-vessel risk, smoking, cancer, blood clots or diverticulitis. Mention pregnancy, breastfeeding, vaccination plans, medicines affecting CYP3A4 and other immunosuppressants. Screening and monitoring include blood counts, liver tests and cholesterol levels.
Can reduce inflammation, symptoms, and flares in eligible patients after appropriate screening and monitoring.
The 45mg regimen is an initial bowel-disease treatment: the reference regimen is eight weeks for ulcerative colitis or twelve weeks for Crohn disease, followed by a lower maintenance strength. Kidney, liver and interaction-related adjustments can change the regimen.
Testing helps identify infection, blood-count, liver, lipid, and safety risks before and during treatment.
No. Splitting, crushing or chewing disrupts the extended-release tablet. A visible tablet remnant in stool does not by itself mean the medicine has failed, but repeated intact remnants should be reported.
Fever, persistent cough, shingles rash, painful urination, wounds, or feeling very unwell should be reported promptly.
Live vaccines are generally avoided during therapy; vaccination should be reviewed before starting.
